Morphology: Monocyte (can differentiate to macrophage)

Culture: Suspension (adherent as macrophage)

Culture Medium: RPMI-1640 with 10% FBS, 1% (2 mM) L-Glutamine, and 1% Pen/Strep. ATCC-recommended addition of 0.05 mM 2-mercaptoethanol is not considered necessary by all labs.

Passaging conditions: Change/add medium as necessary to maintain pH, keeping cells between 200,000 and 800,000 cells per mL. Avoid growing to greater than 1 million cell per mL.

Cryopreservation medium: ATCC recommends pH-equilibrated culture medium + 5% ultra-pure DMSO (or 90% FBS and 10% ultra-pure DMSO)

Suspension cells, do not trypsinize.

Highly sensitive and virtually untransfectable with expression plasmids without virus or nucleofection.

If cells appear to have protrusions/blebs in suspension culture then start new culture from liquid nitrogen. Do not culture for more than 40-60 days from a single culture.

Cells can be differentiated to adherent macrophages by treating with 1-100 nM PMA for 18-36 hours on the final culture substrate. Do not try to trypsinize/scrape and transfer cells after differentiation. Wash cells prior to final experimentation.
